We are currently seeking rising junior and/or senior for a 10-12 week internship during the Summer of 2023.
Objectives/Goals of the Position:
This position will support the development of a rapid, automated Liquid Chromatography Mass Spectrometry (LC-MS) Multi-Attribute Method (MAM) used to monitor the charge variants of Drug Antibody Conjugates (ADCs). Monitoring charge variants for some ADC molecules has been demonstrated to be quite challenging using Ion exchange chromatography (IEC) or Isoelectric focusing (IEF) methods. The reason emanates from the difficulty in decoupling the charge changes due to the payload from the changes to the protein’s post translational modifications (PTMs). The new method would monitor payload conjugation occupancy, and any PTM that results in changes of protein charge.
Primary Duties:
- Evaluating different sample processing methods to reducing sample preparation time.
- Explore different LC gradients to shorten the peptide mapping method.
- Reducing enzymatic digestion time from current 4 hours to one hour or less by evaluating trypsin/Lys C combo digestions, and rapid digestion protocols that use high temperature.
- Evaluating data processing and reporting software to allow for easy, fast, automated data reporting
